At some point between 1600 BC and 1100 BC, King Agrios, son of Portheus and a great-great-grandson of Aetolos (king of Plevron and Calydon), supposedly established Agrinio, an ancient city in the Megali Chora region. The ancient city-states of Aetolia and Acarnania vied for control of the riverbank town, established on the natural boundary between the two states. During the Aetolian League's battles against the Kingdom of Macedonia, Agrinio became a member and was subsequently destroyed by Cassander in 314 BC.

The theatre is situated on a short hill overlooking the Acheloos river near the archaeological site of ancient Stratos, the capital of the Acarnanians. In the late 4th century BC, the theatre had a seating capacity of 6,000 people. Excavation of the stage building's visible foundations has revealed three periods of construction, each dating to the 4th, 3rd, and 2nd centuries BC. This is the biggest of the five theatres discovered in Aetoloacarnania Prefecture so far. Only the foundations of the stage structure, the proscenium, and the parascenium are left, making it a fascinating ruin to explore for history buffs.
